Postdoctoral Position in Malaria: Post-Treatment Hemolysis, Paris, France
Topic outline: Half of the word population is exposed to risk of malaria infection. Artemisinin derivatives are the most efficient and recommended treatment for malaria. Through their mode of action artemisinins induce pitting (expulsion of death parasite by the spleen without lysis of the host red cell). When many pitted red cells are produced during therapy, their delayed clearance few weeks later triggers post-artemisinin delayed hemolysis (PADH). The early quantification of pitted red cells which express P. falciparum HRP2 protein, enables the prediction of PADH. Poor data are available for pitted red cells and how they trigger PADH remains unclear. Using HRP2-based dipsticks test, omics study, immunology and cellular biology, we propose to optimize the test for the prediction of PADH and to characterize pitted red cells in order to determine the mechanism of their elimination. The study will be conducted in French imported severe malaria and in African children (Benin, Mali, Sénégal). This may improve the determination of the incidence of PADH and facilitate the management of millions of patients
Objectives:
The selected candidate will conduct the project to identify and characterize the mechanism of RBC clearance during post-treatment hemolysis follow malaria attack by exploring the role of immune system and determining RBC alterations leading to their elimination.
Qualifications:
- Ph.D. in immunolgy and/or cell & molecular biology
- Expertise in cell-cell interactions and protein fonctions and specially on red cell and or Plasmodium falciparum
- Experience working collaborative networks with multiple partners
- Good communication skills
Start date : Q1 2019
Duration : 30 months funded by the French National Agency for Research (ANR)

L'équipe 4 "Biologie Tissulaire du Globule Rouge" de l'UMR 1134 s'intéresse aux interactions entre les globules rouge et la rate en utilisant des techniques conventionnelles (biochimie, cultures cellulaires, microcopie à fluorescence, cytométrie en flux) et spécifiques (microscopie de contraste interférentiel, ektacytométrie, filtration sur microsphères, Imagestream (Amnis°), perfusion d'organes ex-vivo, électrophysiologie, protéomique) sur des échantillons de laboratoire et des échantillons issus de la recherche clinique. Le contrôle de qualité exercé par la rate sur les globules rouges régule leur microcirculation et les complications au cours de maladies acquises de cette cellule le paludisme, la splénectomie, la thalassémie ou la drépanocytose. L'équipe s'intéresse aussi aux conséquences de ces interactions globule rouge - rate dans le cadre de la transfusion sanguine.
Profil :
Le candidat est titulaire au minimum d'un diplôme de Technicien en biologie ou un profil de type Master 2. Une expérience professionnelle en laboratoire de recherche et si possible dans le domaine de la biologie cellulaire, de l'immunologie et de la biologie moléculaire est fortement souhaitée. Une expérience ou un intérêt pour la recherche clinique et les interactions avec des équipes médicales est un plus.
Principales Fonctions et Missions :
- Participation au projet sur la recherche des mécanismes de clairance parasitaire à la base de l'anémie hémolytique post-artésunate (PADH)
- Gestion de biothèques d'échantillons issus de patients traités pour un paludisme d'importation : préparation de la banque, séparation des fractions du sang total (plasma, globules rouges).
- Mise en œuvre des cultures cellulaires à partir de lignées de plasmodium falciparum ainsi que à partir d'isolats de patients.
- Quantification des protéines parasitaires par méthodes biochimiques
- Etude des mécanismes bio-mécaniques et immunologiques impliquées dans les hémolyses différées post-artésunae.
- Standardisation des techniques et participation à l'analyse des résultats
- Mise à jour des techniques et protocoles du laboratoire
- Participation aux projets de collaboration sur le globule rouge impliquant des déplacements sur site
- Gestion et approvisionnement des stocks de consommables
- Participation active à la démarche qualité, réglementaire et Hygiène et Sécurité
Compétences et qualités requises
- Motivation pour la recherche biologique de niveau international
- Sens de l'initiative et implication forte dans les activités. Une maitrise même modérée de l'anglais est un plus
- Rigueur scientifique, autonomie technique, goût du travail en équipe, sens de l'organisation
- Esprit d'équipe et sens du relationnel
- Maîtrise des applications bio-informatiques du laboratoire
