The "Molecules and Materials Institute of Le Mans" (IMMM) is a joint research unit (UMR 6283) under the supervision of the CNRS and the University of Maine. The Institute includes 60 lecturer-researchers, 12 CNRS researchers, 32 engineers, technicians and administrators, about 55 doctoral students, about thirty Master's students and an annual flow of about ten trainees postdoctoral fellows, visitors and visiting professors. Research activities based on well-established know-how and recognized expertise of chemists, physical-chemistry and physics, fall into four priority scientific themes: organic synthesis, inorganic materials, physics of confined systems and polymers , with different research programs. They thus target bioactive compounds, functional materials having applications in the fields of energy, environment and sustainable development, health, agri-food, plastics, soft matter and optics. State-of-the-art equipment in diffraction, microscopy, local spectroscopy, radiation scattering, rheology, plasma, combined with innovative techniques (ultra-fast spectroscopy pump impulse laser probes), constitute an attractive instrumental platform at the regional, national and international levels. This platform thus constitutes a unique support for a multi-scale and multiscale characterization of innovative materials and nano-functional architectures, complex fluids, and surfaces coupled with recognized expertise in spectral processing and numerical modeling.
Mission (scientific project)
We will work with an original microscopy, allowing to follow the individual trajectories of polymeric particles of a few microns of diameter in interaction with membranes and living cells. In order to study the interactions between colloids and biological membranes, the individual trajectories of colloids at the encounter of membranes and cells of exposed organisms will be followed. The eventual translocation through the membrane barriers is a matter of interest. This project involves collaborative and interdisciplinary work with scientists from a nanoscience institute (IMMM du Mans), bringing expertise in the field of optical techniques, colloids and polymer materials, as well as an institute of biology with an expertise in plastic ecotoxicology in the biological environment (LEMAR member of IUEM, Brest).
General Context: Nanomaterials dispersed in solution are generating considerable effort in many areas, including those related to the environment and health protection. Indeed, because of their small sizes, nanomaterials pose fundamental questions such as their ability to cross membranes thus greatly increasing their potential toxicity. Among these materials, the microplastic and nanoplastic particles (MNP) whose presence in the environment has been recently debated [Ter Halle 2017], raise many concerns including the possible accumulation in the animal and human food chain. Very few experimental approaches make it possible to follow in real time so small particles (submicron and nanometer) during their diffusion in solution. The strong scattering of light by metals or semiconductor particles (quantum-dots) makes them excellent candidates and also explains the large number of studies tracking them. On the other hand, few approaches make it possible to track polymer particles giving incomparably smaller optical signals of equal size. Our main objective is to use microscopies particularly well adapted to visualize the fate of polymers found in a confined and biological space.
Scientific Issues of the Project and Research Program: There is a lack of studies on molecular mechanisms and bio-colloid interactions involving NPMs (The smallest fragments, plastic colloids of less than 20?m or even nanoparticles of a few hundred nanometers, grouped here under the acronym MNP). These are the ones that raise most of the research efforts. For microscopy this comes mainly from their size and weak signals associated in analysis and imaging and for biology this comes from their greater propensity to adsorb on, and / or cross, certain biological membranes [Paul-Pont 2016]. Model migration experiments of MNPs under realistic biological conditions, in interaction with the membranes, will be carried out. Time-resolved optical microscopy will be used (Figure I-A) for reconstruction of trajectories over long distances (hundreds of ?m) and this in-situ. For example, the trajectory of a microparticle of 1 ?m in diameter is shown in Figure I-B. At these acquisition speeds, we distinguish the Brownian shocks undergone by the particle under the effect of the thermal agitation of the solvent, as well as a succession of rebounds on a sliding interface because hydrophobic (located in z = 0).

For the project, we intend to follow the trajectories close to the solid interfaces that will be made biomimetic by functionalizing glass slides [Squillace 2016 and 2017, Figure I-C]. This approach allows to harpoon a lipid membrane floating on a controlled water cushion (in salinity and osmotic pressure), in order to keep it immersed in a fluid and biomimetic state. We aim to monitor diffusion and residence times at the interfaces of MNPs (kinetic and thermodynamic constants), as well as the monitoring of their interaction with membranes. This approach can be deployed on complete cell systems (Figure I-D) in parallel with ecotoxicology tests.
Required Profile
Doctor (PhD) at the physical / biology interface, maximum 3 years of experience after thesis defense . An international experience in research is required (during or after Doctorate). Candidates must not have supported their thesis in the hiring institution and not previously worked in the host research unit. All skills exist in the host team, which allows to maintain a broad but experimental profile and wishing to manipulate biomimetic and / or biological systems and to devote himself to the use of adapted imaging techniques.

L'Institut des Molécules et Matériaux du Mans est une unité mixte de recherche (UMR 6283) sous tutelle du CNRS et de l'Université du Maine. L'Institut des Molécules et Matériaux du Mans (IMMM) comprend 60 enseignants-chercheurs, 12 chercheurs CNRS, 32 ingénieurs, techniciens et administratifs, environ 55 doctorants, une trentaine d'étudiants de Master et un flux annuel d'une dizaine de stagiaires post-doctoraux, visiteurs et professeurs invités. Les activités de recherche qui reposent sur des savoir-faire bien établis et des expertises reconnues de chimistes, physico-chimistes et physiciens, se déclinent selon 4 thématiques scientifiques prioritaires: la synthèse organique, les matériaux inorganiques, la physique des systèmes confinés et les polymères, avec différentes opérations de recherche. Elles visent ainsi des composés bioactifs, des matériaux fonctionnels ayant des applications dans les domaines de l'énergie, de l'environnement et du développement durable, de la santé, de l'agro-alimentaire, de la plasturgie, de la matière molle et de l'optique. Des équipements de pointe en diffraction, microscopies, spectroscopies locales, diffusion de rayonnement, rhéologie, plasma, associés à des techniques innovantes (spectroscopies ultra-rapides pompe sonde lasers impulsionnels), constituent une plateforme instrumentale attractive aux niveaux régional, national et international. Cette plateforme constitue ainsi un support performant pour permettre une caractérisation multiple et multiéchelle des matériaux innovants et des nano-architectures fonctionnelles, des fluides complexes, et des surfaces, couplée à une expertise reconnue dans le traitement des spectres et la modélisation numérique.
Mission (projet)
Ce projet vise l'utilisation d'une microscopie optique, permettant de suivre les trajectoires individuelles de particules polymériques de quelques microns de diamètre en interaction avec des membranes et cellules vivantes. Afin d'étudier les interactions entre colloïdes et membranes biologiques, les trajectoires individuelles de colloïdes à la rencontre de membranes et de cellules d'organismes exposés seront suivies. L'éventuel passage à travers les barrières membranaires est une question d'intérêt. Ce projet engage un travail collaboratif et interdisciplinaire impliquant les chercheurs d'un institut de nanosciences (IMMM du Mans), apportant une expertise dans le domaine des techniques optiques, colloïdes et matériaux polymères, ainsi qu'un institut de biologie dont la thématique des polluants plastiques dans l'environnement biologique est un axe de recherche (LEMAR membre de l'IUEM, Brest).
Contexte général : L'enjeu lié au devenir des nanomatériaux dispersés en solution suscite des efforts considérables dans de nombreux domaines dont ceux liés à l'environnement et la santé. En effet, du fait de leurs petites tailles, les nanomatériaux posent des questions fondamentales comme leur possibilité de traverser des membranes accroissant ainsi fortement leur éventuelle toxicité. Parmi ces matériaux, les particules microplastiques et nanoplastiques (MNP) dont la présence dans l'environnement a été très récemment démontrée [Ter Halle 2017], soulèvent de nombreuses inquiétudes dont la possible accumulation de ces matériaux dans la chaine alimentaire animale et humaine. Très peu d'approches expérimentales permettent de suivre en temps réel des particules de très petite taille (submicronique et nanométrique) dans leur mouvement brownien et pendant leur diffusion en solution. La forte diffusion de la lumière par les particules métalliques ou semi-conductrices (type quantum-dots) en fait d'excellentes candidates et explique aussi le grand nombre d'études les pistant. Peu d'approches permettent par contre de pister les particules polymères donnant des signaux optiques incomparablement plus faibles à taille égale. Notre objectif principal est d'utiliser des microscopies particulièrement bien adaptées pour visualiser le devenir des polymères se retrouvant dans un espace confiné et biologique.
Enjeux scientifiques du projet et Programme de recherche : On constate le manque d'études sur les mécanismes moléculaires et les interactions bio-colloïdales impliquant les MNP. Les plus petits fragments, colloïdes plastiques de moins de 20µm voire nanoparticules de quelques centaines de nanomètres (regroupés ici sous le sigle MNP), sont ceux qui posent le plus de questions à la recherche. Pour la microscopie ceci provient principalement de leur taille et des faibles signaux associés en analyse et en imagerie et pour la biologie ceci provient de leur plus grande propension à s'adsorber sur, et/ou traverser, certaines membranes biologiques [Paul-Pont 2016]. Des expériences modèles de migration des MNP en conditions biologiques réalistes, en interaction avec les membranes, seront réalisées. La microscopie optique résolue en temps sera utilisée (figure I-A) pour reconstruction des trajectoires sur de grandes distances (centaines de µm) et ceci in-situ sur des temps longs. A titre d'exemple, la trajectoire d'une microparticule de 1 µm de diamètre est présentée sur la figure I-B. A ces vitesses d'acquisition, nous distinguons les chocs Browniens subits par la particule sous l'effet de l'agitation thermique du solvant, ainsi qu'une succession de rebonds sur une interface glissante car hydrophobe (située en z=0).

Pour le projet, nous comptons suivre les trajectoires proches des interfaces solides qui seront rendues biomimétiques en fonctionnalisant des lames de verre [Squillace 2016 et 2017, figure I-C]. Cette approche permettant d'harponner une membrane lipidique flottant sur un coussin d'eau contrôlé (en salinité et pression osmotique), afin de la garder en pleine immersion dans un état fluide et biomimétique. Nous visons le suivi de la diffusion et des temps de séjour aux interfaces des MNP (constantes cinétiques et thermodynamiques), ainsi que le suivi de leur interaction avec des membranes. Cette approche pourra se déployer sur des systèmes cellulaires complets (figure I-D) en parallèle de tests écotoxicologiques.
Profil recherché
Docteur déjà à l'interface physique/biologie ou souhaitant y venir avec au maximum 3 ans d'expérience après l'obtention de son doctorat , disposant d'une expérience de recherche à l'international (durant ou après son doctorat) , n'ayant pas soutenu sa thèse au sein de l'établissement de recrutement et n'ayant pas déjà travaillé dans l'unité de recherche d'accueil. Toutes les compétences existent dans l'équipe d'accueil, ce qui permet de conserver un profil large mais expérimental et souhaitant manipuler des systèmes biomimétiques et/ou biologiques et se consacrer à l'utilisation des techniques d'imagerie adaptées.
